Studies of the insects associated with the epidemiological cycle of leishmaniasis on the Caribbean coast of Colombia have been focused on the departments of Sucre and Córdoba. The phlebotomine sand fly fauna of the rest of the Caribbean region remains little known. In the present study four species of the genus Lutzomyia are reported from the department of Cesar for the first time. Sand flies were sampled using two CDC light traps with blue color LEDs at Balneario Hurtado, a tourist area in the city of Valledupar. A total of 50 phlebotomine sand flies were collected, of which the species Lutzomyia cayennensis cayennensis (38 % of the sample) and L. trinidadensis (14 %) were already known from the departament. Specimens of L. evansi (28 %), L. venezuelensis (10 %), L. micropyga (6 %) and L. rangeliana (2 %) represent the first records of these species for Cesar, raising the number of known occurrences from the department to eight species. Noteworthy among the new records is the sandfly L. evansi, recognized vector de Leishmania spp. in the Colombian Caribbean coast.

Mosquitoes are the most important vectors for arboviral human diseases across the world. Diseases such as Dengue Fever (DF), West Nile Virus (WNV), Yellow Fever (YF), Japanese Encephalitis (JE), Venezuelan Equine Encephalitis (VEE), and St. Louis Encephalitis (SLE), among others, have a deep impact in public health. Usually mosquitoes acquire the arboviral infection when they feed on viremic animals (birds or mammals), so their infection can be detected along the year or in short periods of time (seasons). All of this depends on the frequency and seasonality of the encounters between viremic animals and vectors. With the convergence of several phenomena like the increasing traveling of human populations, globalization of economy and more recently the global warming, the introduction of nonendemic arbovirus into new areas has become the current scenario. As examples of this new social and environmental frame we can mention the outbreak of West Nile Virus in North America in the late 1990s and more recently the outbreaks of chikungunya and Zika virus in the Americas. The present chapter deals with one of the first steps in the development of research studies and diagnosis programs, the surveillance of arboviruses in their vectors, the sampling design and mosquito trapping methods. The chapter also includes some important considerations and tips to be taken into account during the mosquito fieldwork.

La toxoplasmosis es una infección producida por el protozoo Toxoplasma gondii. La importancia de la toxoplasmosis reside, principalmente, en la forma congénita y la reactivación en el inmunodeprimido. Cuando la infección es adquirida por primera vez durante el embarazo, puede producir toxoplasmosis congénita, que se presenta en el recién nacido con diversas manifestaciones clínicas, como hidrocefalia, microcefalia y coriorretinitis, o terminar en un aborto. En mujeres gestantes del departamento de Sucre, se ha reportado una alta tasa de seroconversión de infección por T. gondii. Sin embargo, la vigilancia serológica aún no se ha incluido en el control prenatal en embarazadas. El objetivo del presente estudio fue determinar la seroprevalencia de infección por T. gondii en gestantes del departamento de Sucre.

A total of 111 males and 15 females of Trichomyia (Opisthotrichomyia) brevitarsa were captured using CDC light traps in tropical dry forests at the surroundings of the city of Sincelejo, department of Sucre, and Isla Fuerte, department of Bolivar. Collected psychodid flies exhibit all the morphological characteristics ofTr. brevitarsa, including the presence of sensory rods on the first and second palpomere. Successful establishment of a Plasmodium vivax sporozoite challenge model in humans isdescribed. Eighteen healthy adult, malaria-naïve volunteers were randomly allocated to Groups A–C and exposed to 3 ± 1, 6 ± 1, and 9 ± 1 bites of Anopheles albimanus mosquitoes infected with P.vivax, respectively. Seventeen volunteers developed signs and symptoms consistent with malaria,and geometric mean prepatent periods of 11.1 days (9.3–11) for Group A; 10.8 days (9.8–11.9) forGroup B; and 10.6 days (8.7–12.4) for Group C, with no statistically significant difference amonggroups (Kruskal-Wallis, P = 0.70). One volunteer exposed to eight mosquito bites did not developa parasitemia. No differences in parasite density were observed and all individuals successfullyrecovered after anti-malarial treatment. None of the volunteers developed parasite relapses withinan 18-month follow-up. In conclusion, malaria-naive volunteers can be safely and reproducibly infected with bites of 2–10 An. albimanus mosquitoes carrying P. vivax sporozoites. Thischallenge method is suitable for vaccine and anti-malarial drug testing.

A new species of phlebotomine sand ßy, Lutzomyia emberai n. sp, is described and illustrated from specimens collected in a dwelling of the Embera Indian community, situated in the foothills of the Serranõ´a delBaudo´ on the Colombian PaciÞc coast. The morphological characteristics of L. emberai n. sp. suggest that it belongs to the series pia of the group verrucarum, easily differentiated from the other members of this group by diagnostic characters on the palps, labro-pharynx, thorax, and spermathecal ducts. The discovery of this new phlebotomine raises to seven the number of species in the series pia, including Lutzomyia pia (Fairchild & Hertig, 1961); Lutzomyia reclusa Fernandez & Rogers, 1991; Lutzomyia suapiensis Le Pont, Torrez-Espejo & Dujardin, 1997; Lutzomyia tihuiliensis Le Pont, Torrez-Espejo & Dujardin, 1997; Lutzomyia tocaniensis Le Pont, Torrez-Espejo & Dujardin, 1997; Lutzomyia limafalcaoae (Wolff & Galati, 2002); and Lutzomyia emberai Bejarano, Duque & Velez, 2004, n. sp. The taxonomy, distribution, and medicalimportance of this series are reviewed.

Disruption of vacuolar biogenesis in the pathogenic yeast Candida albicans causes profound defects in polarized hyphal growth. However, the precise vacuolar pathways involved in yeast-hypha differentiation have not been determined. Previously we focused on Vps21p, a Rab GTPase involved in directing vacuolar trafficking through the late endosomal prevacuolar compartment (PVC). Herein, we identify two additional Vps21p-related GTPases, Ypt52p and Ypt53p, that colocalize with Vps21p and can suppress the hyphal defects of the vps21 / mutant. Phenotypic analysis of gene deletion strains revealed that loss of both VPS21 and YPT52 causes synthetic defects in endocytic trafficking to the vacuole, as well as delivery of the virulence-associated vacuolar membrane protein Mlt1p from the Golgi compartment. Transcription of all three GTPase-encoding genes is increased under hyphal growth conditions, and overexpression of the transcription factor Ume6p is sufficient to increase the transcription of these genes. While only the vps21 / single mutant has hyphal growth defects, these were greatly exacerbated in a vps21 / ypt52 / double mutant. On the basis of relative expression levels and phenotypic analysis of gene deletion strains, Vps21p is the most important of the three GTPases, followed by Ypt52p, while Ypt53p has an only marginal impact on C. albicans physiology. Finally, disruption of a nonendosomal AP-3-dependent vacuolar trafficking pathway in the vps21 / ypt52 / mutant, further exacerbated the stress and hyphal growth defects. These findings underscore the importance of membrane trafficking through the PVC in sustaining the invasive hyphal growth form of C. albicans.

The purpose of this study was to characterize the effects of azole treatment upon the integrity of the Candida albicans vacuole and to determine whether, in turn, vacuolar trafficking influences azole susceptibility. Profound fragmentation of the C. albicans vacuole can be observed as an early consequence of azole treatment, and it precedes significant growth inhibition. In addition, a C. albicans vps21/ mutant, blocked in membrane trafficking through the late endosomal prevacuolar compartment (PVC), is able to grow significantly more than the wild type in the presence of several azole antifungals under standard susceptibility testing conditions. Sand flies were collected in the municipalities of Puerto Carreño, Cumaribo, and La Primavera, using CDC light traps, Shannon trap, and human bait, and by active search in resting places. The epidemiological importance of these sand fly species is discussed in relation with the cutaneous leishmaniasis.

Trichomyia andina, a new species of non-hematophagous psychodid fly is described and illustrated from males collected in the Risaralda Department, Colombia. This species belongs to the subgenus Opisthotrichomyia Bravo, 2001 and is distinguished from congeners in having a gonostyle broad, multipartite, partially serrated and infuscated, with a long hyaline apex directed inward and backward. The subgenus Opisthotrichomyia is now comprised of seven species, including T. brevitarsa (Rapp 1945), T. vargasi Barretto, 1954, T. nocturna Bravo, 2001, T. festiva Bravo, 2001, T. fluminensis Bravo, 2001, T. riodocensis Alexander, Freitas & Quate, 2001, and T. andina, sp. Nov.

Trichomyia quimbaya sp. nov. is described and illustrated from a male specimen collected with a CDC light trap in the coffee region of the Cordillera Central of Colombia. It can be easily distinguished from others species of Trichomyia by the pair of strong digitiform prolongations of the gonocoxite, both of which end in large spines, with apex directed inwards.

Desde hace más de diez años se registran casos autóctonos de leishmaniasis cutánea y visceral de ocurrencia esporádica en Sincelejo, principalmente en barrios periféricos de esta ciudad, situación que ha estado acompañada por el hallazgo de especies de Lutzomyia (6,14). Sin embargo, se desconoce la variación temporal de estas especies en el entorno urbano; información que podría ser útil para la toma de decisiones en la lucha antivectorial por parte de los entes de salud. Considerando que los flebotomíneos constituyen un riesgo para la población humana como transmisores de Leishmania spp, los objetivos de este trabajo fueron determinar la diversidad de especies del género Lutzomyia y su variación temporal en la zona urbana de Sincelejo.
