Molecular tests are effective for diagnosing acute ZIKV infection, although the majority of infections produce no symptoms at all or present after the narrow window in which molecular diagnostics are dependable. Serology is a reliable method for detecting infections after the viremic period; however, most serological assays have limited specificity due to cross-reactive antibodies elicited by flavivirus infections. Since ZIKV and dengue virus (DENV) widely cocirculate, distinguishing ZIKV infection from DENV infection is particularly important for diagnosing individual cases or for surveillance to coordinate public health responses.

        The objective of the study was to determine the presence of Mycobacterium avium subsp. paratuberculosis (MAP) in cattle faeces from the municipality of Sincelejo, Sucre, Colombia. A direct bacteriological study was performed by microscopy techniques and polymerase chain reaction (PCR) for pools of faeces with two molecular markers, one specific for MAP (IS900) and another based on 16S of the genus Mycobacterium. The presence of acid-fast bacilli occurred in 21.1% of the faecal samples stained with Ziehl Neelsen. Three pools were positive with IS900 and three with 16S. The sequencing indicated the presence of three species of bacteria: MAP, Mycobacterium abscessus and Dietzia timorensis. The presence of MAP in the department of Sucre is identified for the first time.

The dog lived in the urban area of the municipality of Ovejas, Sucre, Colombia. The Indirect Immunofluorescence test was positive for anti-Leishmania spp antibodies up to a dilution of 1/128. The typing of L. infantum was performed by isoenzyme electrophoresis and by 924 Rev Inv Vet Perú 2018; 29(3): 923-930 M. Rivero et al. analysis of the sequence of the cytochrome b gene and the nuclear gene that codes for the heat shock protein of 70 Kda (Hsp70). The present finding is the first confirmation of infection by L. infantum in canines in the urban area of the Colombian Caribbean.

        Among emerging non-albicans Candida species, Candida parapsilosis is of particular concern as a cause of nosocomial bloodstream infections in neonatal and intensive care unit patients. While fluconazole and echinocandins are considered effective treatments for such infections, recent reports of fluconazole and echinocandin resistance in C. parapsilosis indicate a growing problem. The present study describes a novel mechanism of antifungal resistance in this organism affecting susceptibility to azole and echinocandin antifungals in a clinical isolate obtained from a patient with prosthetic valve endocarditis. Transcriptome analysis indicated differential expression of several genes in the resistant isolate, including upregulation of ergosterol biosynthesis pathway genes ERG2, ERG5, ERG6, ERG11, ERG24, ERG25, and UPC2.

        Leptospirosis is one of the most frequent zoonoses worldwide and occurs in tropical, subtropical and temperate areas. The genus Leptospira comprises saprophytic and pathogenic species. The latter isolated from several animals that serve as reservoirs and carriers. The presence of bats in urban areas has increased for various reasons, profusion of plants, lack of predators, presence of luminaries, which has increased the chances of contact between bats, humans and/or pets. The aim of this work was to investigate the presence of pathogenic Leptospira in bats captured in the city of Sincelejo, using the molecular PCR technique. Mist nets were used to capture the bats and these were sacrificed using ether to obtain samples of renal tissue. A fragment of the LipL32 gene was amplified by PCR technique. We identified three families of bats amongst our sample and 26% of them presented pathogenic Leptospira DNA. This represents a great risk to the community in this region.

        Environmental or chemically induced stresses often trigger physiological responses that regulate intracellular pH. As such, the capacity to detect pH changes in real time and within live cells is of fundamental importance to essentially all aspects of biology. In this respect, pHluorin, a pH-sensitive variant of green fluorescent protein, has provided an invaluable tool to detect such responses. Here, we report the adaptation of pHluorin2 (PHL2), a substantially brighter variant of pHluorin, for use with the human fungal pathogen Candida albicans. As well as a cytoplasmic PHL2 indicator, we describe a version that specifically localizes within the fungal vacuole, an acidified subcellular compartment with important functions in nutrient storage and pH homeostasis. In addition, by means of a glycophosphatidylinositolanchored PHL2-fusion protein, we generated a cell surface pH sensor.

        We recently reported that a Candida albicans endosomal trafficking mutant continues to grow after treatment with the azole antifungals. Herein, we report that the vps21Δ/Δ mutant does not have a survival advantage over wild-type isolates after fluconazole treatment in a mouse model of vaginal candidiasis. Furthermore, loss of VPS21 does not synergize with established mechanisms of azole resistance, such as overexpression of efflux pumps or of Erg11p, the target enzyme of the azoles. In summary, although loss of VPS21 function enhances C. albicans survival after azole treatment in vitro, it does not seem to affect azole susceptibility in vivo.

        Although phlebotomine sand flies breeding sites have been identified and recorded by several studies, the microhabitats exploited by these insects remain little-known and hard to find. In this context, the difficulty of finding immature stages, and the limited number of taxonomic studies to identify immature stages of phlebotomine sand flies, are considered the major obstacles when attempting a complete inventory of Lutzomyia species. The objective of this study is to validate Cytochrome Oxidase I (Barcode region) as a marker for the identification of immature stages of Lutzomyia species recovered from natural breeding sites in Colombia. Among 142 collected sand flies, 18 immature individuals that did not complete their life cycle were identified to species level through sequencing of the COI gene. Values of K2P genetic distance between 0.002–0.031 allowed the identification of larvae at species level. The bootstrap support values (96%) in the Neighbor-Joining dendrogram were consistent for the majority of the established MOTUS of Lutzomyia atroclavata, Lutzomyia micropyga, Lutzomyia serrana, Lutzomyia cayennensis, Lutzomyia rangeliana, Lutzomyia shannoni and some species of the genus Brumptomyia. The COI gene is validated as a marker for the identification of immature stages of the genus Lutzomyia.
